Freezing, Handling & Storage of Stallion Semen

The frozen semen course is designed for anyone seriously interested in learning the techniques involved in freezing, thawing, and handling of equine spermatozoa.

Previous experience in semen collection, evaluation, and insemination procedures or attendance at a CSU Equine Reproductive Management equine continuing education short course is required as a prerequisite for this course. Additionally, it is highly recommended to register and complete the online course on Stallion Frozen Semen.

Course Description:  Our two-day course design will allow a more in-depth look at the research we have developed to efficiently freeze equine sperm. Small class size allows for individual attention. The course will emphasize a quality end product and simplify the management of the straws for future use. Participants will have a better understanding of the entire freezing process, quality control, storage, management and artificial insemination and much more. We cover topics in Advanced Reproductive Techniques (ART) such as Intracytoplasmic Sperm Injection (ICSI) and the cryopreservation of epididymal and chemically ejaculated sperm. Hands-on laboratory sessions will provide an opportunity for participants to freeze, thaw and evaluate equine semen. Participants have ample time to work with and become comfortable with all necessary pieces of equipment and products used. Specific lecture and laboratory topics include:

  • Preparation of spermatozoa for freezing & methods for freezing equine sperm
  • Response of spermatozoa to freezing and thawing
  • New freezing extenders
  • Proper handling, storage and thawing procedures
  • Lab equipment requirements
  • Proper use of cushion media and gradients
  • Freezing sperm for ICSI use
  • Refreezing sperm for ICSI use
  • Epididymal sperm recovery, freezing, fertility
  • Chemical ejaculation (for frozen semen)
  • Mare management: conventional frozen/thawed sperm & frozen/thawed epididymal sperm
  • Fertility estimates of frozen semen
