Primers are selected and designed
using Oligo 6 Primer Analysis software (Molecular Biology Insights,
Inc., Cascade Colo.). All primer sets are then determined to be
M. leprae-specific using Blast analysis software from the
Leproma Database Website (http://genolist.pasteur.fr/Leproma/).
Synthesis of these oligonucleotides is performed at the Macromolecular
Resource Facility; a core facility here at Colorado State University.
All oligonucleotides are synthesized and RP-1 purified in house
to ensure that they are essentially free of truncated oligonucleotides,
organic salts, and residual synthesis chemistry. Each oligonucleotide
is lyophilized and stored at -20°C. Reconstitution in 200 μL
of ddH 2O will result in 10 μM stock solution. |
