LSM 510 Meta 405 with Axio Observer Z1 Confocal Microscope Sytem

The Confocal Core unit at the Colorado State University Foothills Campus has a Zeiss LSM 510 META laser scanning confocal microscope is equipped with 4 lasers (one 30mW, Argon =458, 477,488,514 nm; one 10 mW HeNE =543 nm; one 5mW HeNe =633 nm; one 30mW diode =405nm). The automated and reproducible image capture is performed by three photomultipliers (2 standard PMT + 1 META) plus separate PMT for visible light which is recorded simultaneously; and simultaneous interlaced dual laser scanning (multi-tracking: not available in other confocal systems) which permits monitoring simultaneous of 3-label specimens. However, up to 8 different channels can be imaged if it is done sequentially. The LSM 510 scans at 5.2 Frames per second (FPS); 512 x 512, and 77 FPS at 512 x 32 pixels. Line scans at 2800hz. The maximum image size is of 2048x2048 RGB= 12MByte.

Additionally, to permit real-time analysis of biological processes in living cells this configuration has an inverted microscope equipped with a motorized stage and ICS optics, which are unsurpassed for image quality, flexibility and optical perfection. The motorized stage is interchangeable and fully supported by the LSM software. The software automatically identifies the microscope settings and the objectives used and control movements and measurements carried out by the system with high precision. The latter capability allows image capture sufficiently rapidly. For the same purpose this configuration contains a full environmental incubation system that allows heating, as well as temperature, CO2 and humidity controlling devices. Also important for a multi-user friendly facility, the LSM 510 has a robust design with a fast computer analysis and intuitive imaging LSM software, with preset modes for both routine and expert users (which eliminates most manuals adjustments).
