FLOW CYTOMETRY AND CELL SORTING LABORATORY

picture

SERVICES

The Flow Cytometry Laboratory in the Department of Environmental and Radiological Health Sciences at Colorado State University is a multi-user facility equipped with two flow cytometers using Cicero software, including a Coulter EPICS V with two large argon lasers with sorting capabilities. Both machines have tunable wavelength lasers, with an ultraviolet laser option available on the EPICS V.

Flow Cytometers are laser-based cell analyzers which measure various parameters of single cells in suspension. Three PMT's allow for up to three colorfluorescence dye analysis, or one or two color analysis with I90 light scatter. Forward angle light scatter is another parameter, used to measure relative size.

Many possibilities exist for cell analysis, including, but not limited to:
Immunophenotyping (2 - 3 color)
DNA Index for aneuploidy
Cell kinetics (BrdUrd)
Apoptosis
Cell cycle analysis
Intracellular pH
Intracellular free calcium
Intracellular free sodium
Multi-drug resistance
Individual cell sorting
Dead - live cell gating
Mutant isolation
Membrane potential
Membrane fluidity

PERSONNEL

Dr. Mike Fox is the director of the Flow Cytometry and Cell Sorting Laboratory. Dr. Fox graduated from Kansas State University with a Ph.D. in Physics. He has run the flow cytometry facility in the Department of Radiological Health Sciences since 1979. He teaches classes in Radiological Health Sciences and Cell and Molecular Biology. Dr. Fox is active in research involving hyperthermia, DNA analysis and cell kinetics, intracellular ions, heat shock proteins, and tumor studies.
Leslie Armstrong-Lea has been a flow cytometry operator at CSU since 1986 in Dr. Fox's laboratory. Prior to that, she worked in a clinical flow cytometry laboratory at Swedish Hospital in Denver.

SCHEDULING

Dr. Mike Fox will review your experiment with you and give suggestions on how to prepare your cells for the best results. You can reach Dr. Fox at (970) 491-7618, or by email at: Michael.Fox@Colostate.edu.
He is usually available for consultation during your initial experiment as well.
Leslie Armstrong-Lea, the flow cytometry operator, will set up the machine and the parameters needed for your experiment, as well as run your cells through the machine and help interpret your results. Your histograms will be saved to disk for later analysis, if necessary, and you may have printed copies of each histogram as well. After you have established a protocol, further appointment times are available by calling the flow cytometry lab directly at (970) 491-1347, or email Leslie at: Leslie.Armstrong-Lea@Colostate.edu.

RATES

The rates for service for CSU affiliated labs are $85/hour with a one-hour minimum for flow cytometry, with alternate software computer analysis available for $42.50/hour.
Comparable services from a non-CSU vendor are $106.25/hour.
